The technician will provide research support to an immunology laboratory aimed at understanding B cell dysregulation in human disease with an emphasis on autoimmunity in lupus. The work involves isolation of immune cells from human blood and other tissues. The technician will plan and execute experiments under the direct supervision of the PI and/or Sr. Researchers. The technician will use protocols for cell purification, storage, culture, flow cytometry, RNA or DNA extraction, histology, ELISA, and quantitative RT-PCR.
Coordinate experimental plans with team of research clinical coordinators. Coordinators will contact patients and perform sample collection under IRB-approved protocols.
Utilize Biological Specimen Inventory System to enter, track and remove clinical samples from freezers.
Isolate cells of interest from human blood or tissue samples using density centrifugation, magnetic bead sorting, and/or flow cytometry.
Prepare sterile cell culture media, solutions and aliquots.
Set up human cell culture experiments using sterile technique and biochemical safety hood.
Purify RNA and/or DNA from cells or tissue for RT-PCR and bioinformatics experiments
Measure serum and cell culture proteins via ELISA.
Stain samples for flow cytometry.
Utilize flow cytometry core facility for multi-parameter flow cytometric data collection and cell sorting.
Analyze flow cytometry data utilizing Flow Jo software.
Stain tissue sections for histology.
Keep detailed laboratory records of experiments and clearly label samples and reagents.
Create PowerPoint presentations, Excel spreadsheets and graphs/figures of results with Graphpad Prism, R Studio or other similar software package with basic statistical analysis. Assistance is available from other laboratory members or collaborating statisticians if needed.
Participate in lab meetings and relevant seminars, including meetings with the PI and collaborating investigators to propose experiments and review results.
Assist in the management of the laboratory supply inventory.
Oversee equipment maintenance (including centrifuges, incubators, tissue culture hoods, PCR machines, plate reader, and micropipettes) and train individuals in use of equipment.
Ensure compliance with Environmental Health and Safety and Training for Research Laboratory Personnel.
Assist in posters, manuscript and grant preparation.
Other duties may be assigned as needs of the laboratory evolve. Opportunities exist to learn bioinformatic experimental design and analyses including spacial transcriptomics, RNA-seq (bulk and single-cell) and ATAC-seq.
Associate’s degree in appropriate discipline plus 2-3 years specialized experience in related field; or equivalent combination of education and experience. Bachelor’s degree in biology or related field and previous experience with flow cytometry, bioinformatics and/or immunology is preferred. The individual must be dependable and be willing to work in a collaborative team setting.
